New Injection Can Gene-Edit Specific Parts of the Brain, Scientists Say

A team of scientists have developed a new gene-editing tool that, for the first time, can target the specific cells or organs that doctors want edited — including specific areas of the brain.

While testing on lab mice, the Tufts University biomedical engineers developed a gene-editing tool that could specifically target organs like the liver, alter cells in the immune system, and cross the blood-brain barrier that protects the brain in order to edit specific brain cells as well, according to research published in the journal Angewandte Chemie International Edition. And because the tool is directly injected into the bloodstream, it could lead to a straightforward way to perform genetic treatments, should it end up working on people as well.

“We created a method around tailoring the delivery package for a wide range of potential therapeutics, including gene editing,” lead author Qiaobing Xu said in a press release. Depending on the type of cell they want to edit, whether it’s better equipping an immune system cell to fight a certain cancer or a brain cell to treat a neurological condition, Xu and his team slightly modify the delivery package. Glomming different proteins onto the surface, they found, allows the gene-editing enzymes to stick to specific cell types in a way past gene-editing tech wasn’t able to.

“The methods draw upon combinatorial chemistry used by the pharmaceutical industry for designing the drugs themselves, but instead we are applying the approach to designing the components of the delivery vehicle,” Xu added. The system hasn’t yet been tested on human patients. But if it works on them, he said, the system might help doctors treat a wide range of diseases and genetic conditions.

I was lucky enough to do the Working Holiday Visa for a full year, spending six months working in Wellington before visiting Taupo, Rotorua, Hamilton, Auckland, and taking an unforgettable road trip around the South Island before returning to the US. in March 2020, just as the country closed its borders and halted flights. While all visas and applications have been suspended indefinitely due to the COVID-19 pandemic, the below information should help you pounce once the country reopens.

Are you an American under 30? You can probably move to New Zealand for a year

New Zealand’s Working Holiday Visa allows Americans between the ages of 18 and 30 to work in the country for up to 12 months, as well as study or train for up to six months, as long as you fill out an application online and pay a fee of 35 NZD ($23 USD). Once it’s approved, you’ll have 12 months to enter the country. Your visa clock starts when you arrive in New Zealand, so remember to note the date you must leave and set alerts in your calendar to avoid overstaying. Naturally, there are a few catches: You won’t be able to accept any permanent work—this is strictly for casual, temporary, and part-time gigs.

If you have a partner, they’ll have to apply separately, and if you have kids you can’t bring them along. Your passport must be valid for at least 15 months beyond the date you get to New Zealand, so make sure it’s renewed before you start your application if it’s getting close. You’ll also need to have at least $4,200 NZD (about $2,783 USD) in your bank account. While the minimum wage in New Zealand was recently raised to $18.90 NZD ($12.43 USD), you’ll still have to pay for accommodation, transportation, food, and other activities while you’re there, and will need some money to hold you over in case it takes a while to find work.

Man killed in downtown Denver amid dueling 'anti-fascist' and 'patriot' rallies

Just as the “Patriot Muster” appeared to be ending, someone was killed.

A man was killed outside of the Denver Public Library’s downtown branch Saturday afternoon, just as two groups were dispersing after hours of competing ideological rallies in nearby Civic Center Park. The suspect is a private security guard “with no affiliation with Antifa,” the police department tweeted late Saturday. 9News reported the suspect was hired by the television station.

Just after 3:30 p.m. when the shooting happened, the man who was shot fell over and was loaded into an ambulance within a few minutes. Police immediately surrounded the man who appeared to fire the gun and took him into custody. Both the shooter and the victim were white, police say, but they would not give any other information about what brought the victim to the dueling rallies. It was a deadly end to a day that saw a handful of scuffles, plenty of shouting, but little other direct confrontation.

John “Tig” Tiegen, an El Paso County resident who filed re-election paperwork for President Donald Trump in Colorado, a former Marine, and founder of a group called the United American Defense Force, organized what he called a “Patriot Muster.” His group of about 150 people, many of whom were decked out in paramilitary gear and some of whom appeared to be carrying concealed handguns, was gathered in the Greek Amphitheatre at Civic Center Park.

Just to the north side of the theater, the Denver Communists, Colorado Socialist Revolution and other groups held a “Black Lives Matter Anti-Fascist Soup Drive.” They organized it just after Tiegen announced his own event, and collected hundreds of cans of soup through the afternoon. It appeared to be a tongue-in-cheek reference to Trump’s July claim that protesters had assaulted police officers with cans of soup.

Ukraine Deported 2 American Members Of A Neo-Nazi Group Who Tried To Join A Far-Right Military Unit

Ukraine has deported two men from a notoriously violent American neo-Nazi group who tried to set up a local branch and join a far-right military unit to gain combat experience in the war-torn country, according to two Ukrainian security service officials. The men, both US citizens, are members of the neo-Nazi group known as Atomwaffen Division (AWD), one of the Ukrainian officials confirmed to BuzzFeed News. Both officials declined to provide the men’s names and other personal information. It was not immediately clear whether the men were being deported back to the US. A video from the Security Service of Ukraine (SBU) showed an agent alongside the two men with their backpacks in tow at an airport check-in counter.

The deportation of the American neo-Nazis comes as US security officials warn that violent domestic extremists, including white supremacists, pose the greatest terror threat to the US and ahead of next month’s presidential election. During last week's debate, President Donald Trump gave a shoutout to one such group of extremists, the Proud Boys.

AWD is a neo-Nazi group that emerged in 2016 alongside the US alt-right segment of the white supremacist movement, according to the Anti-Defamation League. Members of AWD have already been connected to several killings in the group’s short history, the ADL added. Five senior members were arrested in February and charged with federal crimes. The Southern Poverty Law Center said AWD is “organized as a series of terror cells that work toward civilizational collapse.”

The SBU said in a statement on its website Thursday that the two Americans “produced a video promoting neo-Nazism and urging citizens to commit particularly serious crimes, including murder and terrorist attacks” in Ukraine. The SBU said the video, screenshots of which it shared online and showed the “Atom” in Atomwaffen, was spread across social networks. BuzzFeed News has viewed the video, which was originally published in 2019. It showed several men in masks and camouflage fatigues donning what appear to be assault rifles. The Americans carried out “illegal activities” in the capital city, Kyiv, as well as the western city of Lviv and the eastern city of Kharkiv, the SBU said. Moreover, it said, they “tried to join one of the Ukrainian military units in order to gain combat experience, which the representatives of the group planned to use in illegal activities.”

The 2020 General Election on November 3 may still feel like a ways off, but considering the pace at which 2020 has mercifully flown by so far, it’ll be here before you know it. And what a relief that will be. Hopefully. But that also means the important deadlines you’ll need to meet in order to cast your ballot might sneak up on you, too. Don’t freak out. Now’s the time to do some research and make a plan for how you’ll vote -- and vote safely -- in this election to ensure your voice is heard. We’re here to make that a little easier with this comprehensive guide to voting in all 50 states and the District of Columbia.

Voting is probably going to be a bit different this year due to safety concerns related to the coronavirus pandemic, and of course, the resulting focus on voting by mail in many states. For a lot of voters, the experience won’t resemble the usual process of heading over to a polling place, walking into a voting booth, and casting a ballot, though that’s certainly still an option. Should you choose to take that route, there are several ways you can reduce your risk of getting or spreading COVID-19. The key is to find out what options are available in your state and to figure out which one is best for you.

Of course, casting a ballot is easier in some states than in others. Every state has its own set of ways to vote, deadlines, and requirements. In fact, early voting -- whether it be the traditional in-person way or via absentee ballots -- has already started in Illinois, Minnesota, Michigan, Pennsylvania, New Jersey, and more as of late September, and voters are receiving mail-in ballots in many other places across the country. The election has already begun. There’s a chance you could cast your ballot today and get it all out of the way should you choose.

REPUBLICANS HAVE A HISTORY OF CHEATING

Republicans are masters of voter suppression, having spent decades restricting the vote and voting power of those who tend to vote Democratic. A small sampling of their nefarious efforts:

Since 2013, states across the South have closed more than 1,200 polling sites; more than half of those lost were in Texas and seven counties in Georgia have been left with only a single polling location.
Between 2011 and 2016, Ohio purged more than 2 million voters from its rolls, with Black voters twice as likely to be removed compared to white voters, while in one night in 2017 Georgia purged 500,000 voters.
In 2016, gerrymandering led to Republicans winning 22 more seats in the U.S. House; across the country, four times as many states had Republican-skewed state Assembly and House districts than those that favored Democrats.

Trump Fatigue Is Finally Here

And the timing couldn’t be better.

WASHINGTON, DC -- We have no idea what Donald Trump’s physical condition is. We have no idea the extent of his illness or whether he’ll recover. We have no idea when he contracted COVID. We have no idea when his last negative test was administered. We have no idea whether he’s going to do something erratic as a consequence of the medication he’s taking. We know nothing for certain, other than he’s positive for COVID and the course of the infection was bad enough to land him in Walter Reed for a long weekend. That’s it. There’s really only one way to at least have a sense of what’s happening: listen to what Trump and his henchmen say publicly and then assume the opposite is true. In most cases, this is at least functional in terms of knowing what’s real and what’s fiction. For example, when Trump says he’s feeling great, he’s probably lying -- he’s probably not feeling well, given the virus, and the only reason he seems somewhat together is likely a side effect of the steroids.

It’s this guessing game, however, that’s beginning to wear thin with voters -- this ongoing round of grabass in which we’re left with no choice but to unravel Trump’s geyser of gibberish as if we’re trying to pull chewing gum out of our hair. Only, as soon as we get frustrated and just cut the damn hair off, he jams another wad of Hubba Bubba in there, and around we go again. It simply never ends. Between the hasty roll-out of Amy Coney Barrett, then Trump’s horrifying performance at the debate, and then the COVID diagnosis accompanied by more incompetence and deception, all layered precariously on top of all of the other things including 210,000 dead, American voters seem like they’re done with the horseshit. This is the dreaded Trump fatigue. It’s finally here and the timing couldn’t be better. Of course, I cut all of this with my standard warning: don’t get happy. In other words, don’t get complacent. Don’t for a second believe this is over.

There are still 26 days to go, and as you’re well aware by now, 12 different catastrophes can drop in our laps within the next 26 seconds, much less 26 days. And that includes a ton of cheating and foreign cyber-attacks. This is not over. But if the election were held today, and all things were normal, we’re looking at a result for Joe Biden that’s in line with Barack Obama’s 2008 victory -- and possibly bigger than that. If you recall, Obama walked away with 365 electoral votes, and previously unheard-of wins in states like North Carolina, Iowa, and even Mike Pence’s Indiana. According to RCP’s “no toss-up” map, Joe Biden is on the verge of winning 374 electoral votes, nine more than he and Obama won in 2008. While the map doesn’t include Indiana in the blue column, if the election were today, Biden would win the normally red states of Arizona and Georgia -- the entire east coast minus South Carolina. Now, bear in mind, this is a stylized map in which RCP awards each state based on even the narrowest margins in the polls, disregarding the margins of error

Nevertheless, the polls are moving heavily in the direction of Biden and Kamala Harris at the moment, eating into the ranks of the Red Hats -- some of Trump’s hardcore supporters are starting to peel off from the collective. This has to be Trump fatigue. The way this manifests itself among his supporters, though, is different than how it impacts Normals, be they political junkies or average Americans. Trump supporters might be just plain sick of defending their guy when their guy seems to perpetually make things worse for himself. It becomes exhausting when the targets keep moving around -- Trump contradicting himself, or doing blatantly indefensible things like throwing off his mask upon arriving at the White House after being in the hospital for an incurable infectious disease. So, things are looking brighter than they did even several days ago. Personally, I’m relieved that there doesn’t appear to be a sympathy vote showing in the polls. Americans are done with Trump and their sympathies are being dedicated to people who deserve it.
