Of blood coming out of a body without a cut or injury. It freaked them out.
Centuries ago, they figured out enough they could relate a period with pregnancy, but really didn't understand the hows or whys until science and could advance far enough that a woman wouldn't die while they were trying to figure out the actual way the reproductive system worked.

Isolation of women during their period made sense if a culture believed the male sexual organs could possibly be affected through some sort of infection or poisoning process if a grown man touched a woman bleeding from her sexual organs. But now-a-days, it's just a lingering superstitious practice.
