There are plausible estimates that the pre-Columbian population of ALL of the Americas (North, South, Central and the Caribbean) may have been around 100 million people before first contact. It's estimated that the populations dropped between 50% and 90% in the 1500's as diseases shot across the America's following that contact, simply due to the native populations trading with each other along their own trade routes. When the first European colonists showed up many decades later to claim some of that land, they found the land much emptier than it had been a generation before.
It's estimated that the largest Native American "cities" had populations up around 100,000 people. That's about as big as you can get without wheels.
